| I. Background | |

4-1BB [也稱(chēng)為 CD137 或 TNF 受體超家族成員 9 (TNFRSF9)] ，是糖基化的 I 型膜蛋白，屬于 TNFR 超家族成員，由四個(gè)胞外富含半胱氨酸的假重復結構域組成。4-1BB 的胞質(zhì)區包含 TNF 受體相關(guān)因子 (TRAF) 結合基序。4-1BB 表達主要發(fā)現于活化的細胞毒性CD8 + T 細胞和輔助性 CD4 + T 細胞的表面，活化后，NK 細胞、B 細胞、單核細胞和樹(shù)突狀細胞中可誘導 4-1BB 表達。作為可誘導的共刺激分子，4-1BB 可增強 T 細胞對抗原的反應。在 T 細胞上被 4-1BBL 上調和三聚化后，4-1BB 會(huì )募集 TRAF 銜接蛋白至細胞漿TRAF 結合基序，從而啟動(dòng)共刺激信號傳導。
4-1BBL (TNFSF9) 是 4-1BB 的主要配體，主要在 APC 、B 細胞、巨噬細胞和其他細胞類(lèi)型中表達，并與 4-1BB 結合，通過(guò) TRAF1 和 TRAF2 誘導信號傳導，從而激活 NF-κB、AKT、p38 MAPK 和 ERK 通路。NF-κB 誘導 BCL-XL、BCL-2 和 c-FLIP 的轉錄，并刺激IL-2、IL-4 和 IFN-γ 的表達，從而促進(jìn) T 細胞的存活和細胞毒性活性。因此，4-1BB/4- 1BBL 信號通路可以促進(jìn)不同類(lèi)型的細胞存活。
