| I. Background | |
| 趨化因子是一類(lèi)炎性細胞因子，具有調節細胞遷移、粘附、吞噬作用、細胞因子分泌、 增殖和細胞凋亡的能力。越來(lái)越多的證據表明，趨化因子及其受體在穩態(tài)和炎癥條件下在 調節樹(shù)突狀細胞遷移以及穩態(tài)和炎癥功能方面發(fā)揮著(zhù)重要作用。 CXCR2（C-X-C 趨化因子受體 2）是 G 蛋白偶聯(lián)受體（GPCR）家族成員，屬于 CXC 趨化因子受體亞家族，廣泛參與中性粒細胞趨化、炎癥反應及腫瘤微環(huán)境調控。CXCR2 通 過(guò)結合多種趨化因子（如 CXCL1、CXCL2、CXCL8/IL-8）激活下游信號，在免疫應答、 組織修復及病理過(guò)程中發(fā)揮關(guān)鍵作用。相較于 CXCR1，CXCR2 配體結合范圍更廣（包括 CXCL1-3、CXCL5-8），且在炎癥性疾病和癌癥中表現出更強的促血管生成及免疫抑制特 性。 |

| II. Description | |
CXCR2 β-Arrestin CHO 報告基因藥靶模型很好的模擬了體內 CXCR2/β-Arrestin 的信號 轉導過(guò)程，原理見(jiàn)下圖所示。當缺乏配體刺激時(shí)，β-Arrestin 不與 CXCR2 結合，融合β- Arrestin 的熒光素酶處于失活構象，當 CXCR2 遭遇配體刺激時(shí)，融合熒光素酶報告基因的 β-Arrestin 被招募，使熒光素酶報告基因處于激活狀態(tài)，加入其底物后發(fā)光信號增強。 Figure 1. CXCR2 β-Arrestin CHO 細胞模型原理圖 |
