| I. Background | |
| CLDN1 基因位于人類(lèi)染色體 3q28.2，屬于緊密連接蛋白家族（Claudins）成員。 CLDN1 基因編碼的緊密連接蛋白 Claudin-1 為四次跨膜蛋白，包含兩個(gè)細胞外環(huán)（ECL1、 ECL2）和 N/C 端胞內尾。ECL1 通過(guò)二硫鍵穩定結構，參與細胞旁通道調控；C 端與胞內 支架蛋白結合，介導信號傳導。 在上皮細胞和內皮細胞中，Claudin-1 通過(guò)同型/異型相互作用形成緊密連接，調控細胞 旁通透性。而 Claudin-1 與胞內蛋白（如 Zonula Occludens-1）結合，參與 Wnt/β-catenin、 TGF-β等通路，影響細胞增殖、分化及遷移。在腫瘤中，其異常表達可激活促癌信號。 Claudin-1 作為緊密連接的核心組分，其功能貫穿屏障維持、信號調控及疾病發(fā)生。隨 著(zhù)對其機制的深入解析，針對 Claudin-1 的靶向治療在腫瘤、慢性炎癥及退行性疾病中展現 出廣闊前景。 |
