| I. Background | |
| 白細胞介素 23 （IL23） 是慢性炎癥性疾?。ㄈ玢y屑病、炎癥性腸病、多發(fā)性硬化癥 或類(lèi)風(fēng)濕性關(guān)節炎）發(fā)展的關(guān)鍵促炎細胞因子，與 IL12、IL27、IL35 和 IL39 一起屬于 IL12 家族 。過(guò)度 IL23 信號轉導的病理后果與其促進(jìn)靶細胞群（主要是分泌 Th17 和 IL17 的 TCRγδ 細胞 （Tγδ17） ）產(chǎn)生炎癥介質(zhì)的能力有關(guān)，例如 IL17、IL22、粒細胞-巨噬細 胞集落刺激 （GM-CSF） 或腫瘤壞死因子 （TNFα）。這些致病介質(zhì)促進(jìn)粒細胞和巨噬細 胞的募集和活化，導致組織損傷，誘發(fā)慢性炎癥，最終發(fā)展出臨床癥狀。 IL23 是一種異二聚體細胞因子，由兩個(gè)二硫鍵連接的結合亞基 p40（由 IL12β編碼） 和 p19 （IL23α） 形成 ，它們與 IL12Rβ1 和獨特的 IL23R 形成的膜受體復合物結合。IL23 與 IL12 共享 p40 亞基及其受體成分之一 IL12Rβ1。盡管存在相同的亞基，但 IL23 和 IL12 具有不同的生物學(xué)功能。IL23 刺激 JAK2 和 TYK2 活性，導致信號轉導和轉錄激活因子 （STAT）的激活。 |
